Tato část ISO 10705 určuje metodu pro průkaz přítomnosti a kvantitativní stanovení somatických kolifágů pomocí inkubace vzorku s příslušným hostitelským kmenem. Metodu lze použít pro všechny druhy vod, sedimentů a extraktů kalů, pokud je nutné, pak po zředění. Metoda je použitelná také pro extrakty z jedlých mořských měkkýšů (ústřic), eventuálně korýšů. V případě malého počtu fágů může být nezbytné zkoncentrování; pro tento postup bude zpracována samostatná část ISO 10705.
