Tato česká norma specifikuje metodu stanovení počtu kontaminujících mikroorganismů technikou počítání kolonií vykultivovaných při 30 °C. Metoda je použitelná pro máslo, kysaná mléka a čerstvé sýry. V normě se aplikuje technika očkování na povrch zde specifikované agarové půdy (5.6), jejíž všechny musí být prosté sacharidů. Naočkované plotny se inkubují aerobně při 30 °C _ 1 °C po dobu 72 h.
